GW501516 acts as a ligand for PPAR delta, a nuclear receptor that regulates transcription of genes involved in fatty acid oxidation and energy use. Activation of this receptor in skeletal muscle shifts metabolism toward fat burning in animal models. The compound does not burn fat directly; it changes gene expression over hours to days. Researchers study it to understand metabolic flexibility and exercise adaptation. Effects observed in rodents are not automatically expected in humans.
Preclinical research reported that GW501516 increased running endurance in mice and improved lipid profiles in some animal species. Early human trials explored effects on high-density lipoprotein cholesterol, triglycerides, and glucose handling, but the program was discontinued. Published human data are sparse and do not establish efficacy for any condition. Studies also examined PPAR delta in cancer biology, with conflicting findings across models. The relationship between receptor activation, tissue context, and disease risk remains an active area of investigation.
Anti-doping laboratories identify GW501516 and its metabolites using liquid chromatography-tandem mass spectrometry. Urine is the usual matrix, and detection can occur after the parent compound has cleared from blood. The exact detection window depends on dose, formulation, individual metabolism, and assay sensitivity. Because the compound is prohibited at all times, athletes are subject to testing in and out of competition. Analytical methods continue to improve as new metabolites and designer analogs are characterized.
Anti-doping laboratories detect GW501516 and its metabolites using liquid chromatography-tandem mass spectrometry. Urine is the most common matrix, though blood and dried blood spots may also be used in some programs. Detection depends on factors such as dose, timing, metabolism, and the sensitivity of the assay. Published methods describe limits of detection in the low nanogram per milliliter range for related compounds. Exact detection windows are not fixed for all situations and remain an area of ongoing study.
Products sold as cardarine have been found to contain incorrect compounds, variable amounts, or no active ingredient at all. Independent testing is required to verify identity and purity. Common analytical approaches include high-performance liquid chromatography, mass spectrometry, and nuclear magnetic resonance for structural confirmation. These methods can distinguish GW501516 from related PPAR agonists and from unrelated steroids. For regulators and researchers, such verification is central to interpreting both biological results and adverse event reports.
Cardarine is prohibited in competitive sport under the World Anti-Doping Agency code, where it is classified as a metabolic modulator. It is not approved as a prescription medicine in the United States, European Union, or other major markets. Regulatory action has focused on its presence in sports and in products marketed as research chemicals. Because it has no accepted medical indication, supply is often unregulated. This status creates legal and safety uncertainties for anyone who encounters the substance.

== Mechanism == The basis of many functional gastrointestinal disorders is distension of the intestinal lumen. Such luminal distension may induce pain, a sensation of bloating, abdominal distension and motility disorders. Therapeutic approaches seek to reduce factors that lead to distension, particularly of the distal small and proximal large intestine. Food substances that can induce distension are those that are poorly absorbed in the proximal small intestine, osmotically active, and fermented by intestinal bacteria with hydrogen (as opposed to methane) production. The small molecule FODMAPs exhibit these characteristics. Ingestion of certain short-chain carbohydrates, including lactose, fructose and sorbitol, fructans and galactooligosaccharides, can induce gastrointestinal discomfort similar to that seen in IBS. Dietary restriction of short-chain carbohydrates is associated with improvement of symptoms. These short-chain carbohydrates (lactose, fructose and sorbitol, fructans and GOS) behave similarly in the intestine. Firstly, being small molecules and either poorly absorbed or not absorbed at all, they drag water into the intestine via osmosis. Secondly, these molecules are readily fermented by colonic bacteria, so upon malabsorption in the small intestine they enter the large intestine where they generate gases (hydrogen, carbon dioxide and methane). The dual actions of these carbohydrates cause an expansion in volume of intestinal contents, which stretches the intestinal wall and stimulates nerves in the gut.
=== Biochemistry === In proteomics, MALDI is used for the rapid identification of proteins isolated by using gel electrophoresis: SDS-PAGE, size exclusion chromatography, affinity chromatography, strong/weak ion exchange, isotope coded protein labeling (ICPL), and two-dimensional gel electrophoresis. Peptide mass fingerprinting is the most popular analytical application of MALDI-TOF mass spectrometers. MALDI TOF/TOF mass spectrometers are used to reveal amino acid sequence of peptides using post-source decay or high energy collision-induced dissociation (further use see mass spectrometry). MALDI-TOF have been used to characterise post-translational modifications. For example, it has been widely applied to study protein methylation and demethylation. However, care must be taken when studying post-translational modifications by MALDI-TOF. For example, it has been reported that loss of sialic acid has been identified in papers when dihydroxybenzoic acid (DHB) has been used as a matrix for MALDI MS analysis of glycosylated peptides. Using sinapinic acid, 4-HCCA and DHB as matrices, S. Martin studied loss of sialic acid in glycosylated peptides by metastable decay in MALDI/TOF in linear mode and reflector mode. A group at Shimadzu Corporation derivatized the sialic acid by an amidation reaction as a way to improve detection sensitivity and also demonstrated that ionic liquid matrix reduces a loss of sialic acid during MALDI/TOF MS analysis of sialylated oligosaccharides.
Frederick Sanger (; 13 August 1918 – 19 November 2013) was a British biochemist who received the Nobel Prize in Chemistry twice. He won the 1958 Chemistry Prize for determining the amino acid sequence of insulin and numerous other proteins, demonstrating in the process that each had a unique, definite structure; this was a foundational discovery for the central dogma of molecular biology. At the newly constructed Laboratory of Molecular Biology in Cambridge, he developed and subsequently refined the first-ever DNA sequencing technique, which vastly expanded the number of feasible experiments in molecular biology and remains in widespread use today. The breakthrough earned him the 1980 Nobel Prize in Chemistry, which he shared with Walter Gilbert and Paul Berg. He is one of only three people to have won multiple Nobel Prizes in the same category (the others being John Bardeen in physics and Karl Barry Sharpless in chemistry), and one of five persons with two Nobel Prizes.
On 15 April 1944, VIII Fighter Command began "Operation Jackpot", attacks on Luftwaffe fighter airfields. As the efficacy of these missions increased, the number of fighters at the German airbases fell to the point where they were no longer considered worthwhile targets. On 21 May, targets were expanded to include railways, locomotives, and other rolling stock used by the Germans to transport materiel and troops, in missions dubbed "Chattanooga". The P-51 excelled at this mission, although losses were much higher on strafing missions than in air-to-air combat, partially because the Mustang's Merlin engine, being liquid cooled, was vulnerable to radiator and coolant line damage from small-arms gunfire. On the other hand, the Mustang's stablemate, the Republic P-47 Thunderbolt, being powered by an air-cooled radial engine, could usually shrug off small-arms fire, and thus was regularly tasked with ground-strafing missions.

=== Insulin access and innovation === In response to rising insulin costs and research showing that many people with diabetes are skipping or rationing their insulin, Breakthrough T1D has advocated for insurance companies to provide better health coverage for those living with type 1 diabetes, including making out-of-pocket costs for insulin and other vital diabetes tools more predictable and reasonable. The aim of this advocacy is to ensure people have the freedom to choose treatment strategies that are appropriate for them, and to cover artificial pancreas/automated insulin delivery systems. In response to escalating insulin affordability issues, Breakthrough T1D partnered with nonprofit drug maker Civica to manufacture insulin that will cost $30 a vial, regardless of a patient's insurance provider. Civica insulin is expected to be available to the public in 2025. Breakthrough T1D also advocates for passage of the INSULIN Act, which caps cost-sharing under private health insurance for a month's supply of selected insulin products at $35 or 25% of a plan's negotiated price (after any price concessions), whichever is less, beginning in 2025.
Still, during the Soviet era, Jews were encouraged to admire Cossacks as the antitheses of the "parasitic" and "feeble dwellers of the shtetl." A number of Yiddish writers, including Khaim Melamud, Shmuel Gordon, Viktor Fink, and Shmuel Godiner, presented fictionalized accounts of peaceful Jewish-Cossack coexistence, while efforts were made by the pro-Soviet press to present Khmelnytsky as a heroic figure and Cossacks as liberators from the Nazis. Historiography interprets Cossackdom in imperial and colonial terms. In Ukraine, where Cossackdom represents historical and cultural heritage, some people have begun attempting to recreate the images of Ukrainian Cossacks. Traditional Ukrainian culture is often tied in with the Cossacks, and the Ukrainian government actively supports these attempts. The traditional Cossack bulava serves as a symbol of the Ukrainian presidency, and the island of Khortytsia, the origin and center of the Zaporozhian Sich, has been restored. The video game Cossacks: European Wars is a Ukrainian-made game series influenced by Cossack culture. Cossacks are also mentioned outside Europe. The Japanese anime The Doraemons, part of the larger Doraemon anime series, has a Cossack character, Dora-nichov, who is from Russia.
Aeroponics is also widely used in laboratory studies of plant physiology and plant pathology. Aeroponic techniques have been given special attention from NASA since a mist is easier to handle than a liquid in a zero-gravity environment.
In the dominant post-war narrative of West Germany, the Nazi regime was characterised as having been a 'criminal' state, illegal and illegitimate from the outset; while the Weimar Republic was characterised as having been a 'failed' state, whose inherent institutional and constitutional flaws had been exploited by Hitler in his illegal seizure of dictatorial powers. Consequently, following the death of Hitler in 1945 and the subsequent capitulation of the German Armed Forces, the national political, judicial, administrative, and constitutional instruments of both Nazi Germany and the Weimar Republic were understood as entirely defunct, such that a new West Germany could be established in a condition of constitutional nullity. Nevertheless, the new West Germany asserted its fundamental continuity with the 'overall' German state that was held to have embodied the unified German people since the Frankfurt Parliament of 1848, and which from 1871 had been represented within the German Reich; albeit that this overall state had become effectively dormant long before 8 May 1945. In 1949 with the continuation and aggravation of the Cold War (for example, the Berlin Airlift of 1948–49), the two German states that had originated in the Western Allied and the Soviet Zones respectively became known internationally as West Germany and East Germany. Commonly known in English as East Germany, the former Soviet occupation zone in Germany, eventually became the German Democratic Republic or GDR.

=== Intein-containing proteins === An intein is a protein contained within another protein, the extein. Parasitic DNA infects an intein gene, which encodes an endonuclease. The resulting cDNA (complementary DNA) encodes the extein along with the intein. The intein contains a self-cleaving domain, which has the endonuclease nested within it. The intein domain performs two proteolytic cleavages at its own N-terminus and C-terminus and releases from the extein, separating it in two fragments. This two fragments are then spliced together and the extein remains as a completely functional protein. The N-terminal residue of the intein domain must be a serine, threonine or cysteine, and it attacks its preceding peptide bond in order to form an ester or a thioester. The first residue of the second portion of the extein must be a serine, threonine or cysteine as well, and this second nucleophile forms a branched intermediary. The C-terminal residue of the intein domain is always an asparagine, which cyclizes to form a succinimide, cleaving its own peptide bond and releasing the intein from the extein. Finally, in the extein the ester or thioester bond is rearranged to form a normal peptide bond. There are three known families of intein-containing proteins (N9, N10 and N11) all of them included in the PD clan, which contains proteolytic enzymes of different catalytic types. The tertiary structure has been solved for the intein V type proton ATPase catalytic subunit (Saccharomyces cerevisiae), a member of family N9 and for several inteins from family N10.
Some historians assert that as many as 17 million people were sold into slavery on the coast of the Indian Ocean, the Middle East, and North Africa, and approximately 5 million African slaves were bought by Muslim slave traders and taken from Africa across the Red Sea, Indian Ocean, and Sahara Desert between 1500 and 1900. The captives were sold throughout the Middle East. This trade accelerated as superior ships led to more trade and greater demand for labour on plantations in the region. Eventually, tens of thousands of captives were being taken every year. The Indian Ocean slave trade was multi-directional and changed over time. To meet the demand for menial labour, Bantu slaves bought by east African slave traders from southeastern Africa were sold in cumulatively large numbers over the centuries to customers in Egypt, Arabia, the Persian Gulf, India, European colonies in the Far East, the Indian Ocean islands, Ethiopia, Sudan and Somalia. According to the Encyclopedia of African History, "It is estimated that by the 1890s the largest slave population of the world, about 2 million people, was concentrated in the territories of the Sokoto Caliphate. The use of slave labour was extensive, especially in agriculture." The Anti-Slavery Society estimated there were 2 million slaves in Ethiopia in the early 1930s out of an estimated population of 8 to 16 million. Slave labour in East Africa was drawn from the Zanj, Bantu peoples that lived along the East African coast.
==== Case study ==== The Kennewick Man is a notable example of human remains caught in a struggle between scientific merit and cultural traditions. Since his discovery in 1996, his fate has been the topic of great controversy. As one of the oldest well-preserved ancient skeletons found in America, scientists are eager to conduct various testing on the remains. Native American groups, however, have been adamantly calling for his repatriation and reburial, as per their traditions.
Dried fruit Soft drinks Juice Sugar as sweets, tablets or cubes. The anti-hypertensive class of medication known as calcium channel blockers could be useful for reactive hypoglycemia as inhibition of the calcium channels on beta islet cells can help prevent an overproduction of insulin after a meal is eaten.

It binds to and activates PPAR delta, a nuclear receptor that controls expression of genes related to fatty acid oxidation. This mechanism can alter energy metabolism in animal models. It is not a direct stimulant or fat-burning enzyme.
Early-stage trials examined lipid and glucose markers, but the development program was discontinued. Published human results are limited and do not support approved use for any indication. Claims of performance or health benefits remain unproven.
Yes. Laboratories use LC-MS/MS to detect GW501516 and its metabolites in urine. Detection depends on timing and sensitivity, but the substance is banned at all times.
Legal status varies by country, but cardarine is not approved as a medicine in major jurisdictions. It is often sold as a research chemical, a category that may not be lawful for human use. Buyers should check local laws and product labels carefully.
